Would a podcast producer brother of the Secretary of Defense, holding the position of lead advisor to the Secretary of Defense be considered a DEI hire? I thought positions were based on merit and qualifications, no?

They're not laying people off because they're DEI hires. They're laying them off because they make the federal government and its agencies work, which is not what Trump and his backers want.

They weren't fired for being bad at their jobs, or for being unqualified in some way. They were laid off en masse to gut the Federal Government's ability to do its job.
