I have a lesbian friend who got hit on by a gay guy because he thought she was a dude. We all had a good laugh about that one.

Unless you have a romantic or sexual interest in the person, why really care what gender they are?

Actually there are cases where people are born with both male and female parts. It's rare but it happens.
